{"id":56318,"date":"2018-11-13T11:58:41","date_gmt":"2018-11-13T10:58:41","guid":{"rendered":"https:\/\/biovet-alquermes.com\/effect-of-immunostimulant-pronutrients-in-macrophages\/"},"modified":"2024-04-22T12:10:58","modified_gmt":"2024-04-22T10:10:58","slug":"effect-of-immunostimulant-pronutrients-in-macrophages","status":"publish","type":"post","link":"https:\/\/biovet-alquermes.com\/en\/2018\/11\/13\/effect-of-immunostimulant-pronutrients-in-macrophages\/","title":{"rendered":"Effect of immunostimulant pronutrients in macrophages"},"content":{"rendered":"<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone wp-image-9035 size-full\" src=\"https:\/\/biovet-alquermes.com\/wp-content\/uploads\/2024\/04\/511319cb2876b9d78f25ea822cfafc14935eae9c-1-2.jpg\" alt=\"\" width=\"900\" height=\"600\" \/><\/p>\n<h5>INTRODUCTION<\/h5>\n<p>An in vitro test, carried out in a private laboratory in Spain in 2015, researched the <strong>molecular mechanisms of the immunostimulant effect of a natural product rich in pronutrients.<\/strong><\/p>\n<p><strong>Pronutrients are molecules of botanical origin<\/strong> that produce the activation of a target cell, which in this case are macrophages, and induce an increase in the translation of RNA to proteins.<\/p>\n<p>In order to calculate the effect of pronutrients on the mRNA-protein translation rate, a technique called SUnSET (Surface Sensing of Translation) was used.<\/p>\n<p><strong>SUnSET technique is a non-radioactive method for monitoring protein synthesis in cell populations,<\/strong> based on the use of puromycin. Puromycin is a tyrosyl-tRNA analogue that can join newly synthesized proteins by a peptide bond, so the detection of puromycin by an anti-puromycin antibody (a technique called Western Blot-ELISA) can be correlated with the translation rate mRNA-protein.<\/p>\n<p><em><img loading=\"lazy\" decoding=\"async\" class=\"alignnone wp-image-9036 size-full\" src=\"https:\/\/biovet-alquermes.com\/wp-content\/uploads\/2024\/04\/IMAGEN-INMUPLUS-RECOMBINA-300x294-1-2.png\" alt=\"\" width=\"300\" height=\"294\" \/><\/em><\/p>\n<p><em>Picture 1: Protein synthesis and puromycin incorporated to a protein.<\/em><\/p>\n<h5>METHODOLOGY<\/h5>\n<p>In vitro assays were performed in 12 wells, where 1.2 \u00d7 105 cells\/ml were added in a final volume of 2 ml per well.<\/p>\n<p>After 4 days of culture, confluence was observed, and the medium was replaced with different supplements, following the scheme described below:<\/p>\n<p>ASSAY A (Control): Only puromycin is added.<br \/>\nASSAY B (Samples): Puromycin and the natural immunostimulant are added.<br \/>\nASSAY C (Control). Puromycin and cycloheximide are added.
